Transcription factors (TFs) are proteins that control the rate of transcription of genetic information from DNA to messenger RNA. They do this by binding to specific DNA sequences near genes (often at promoter or enhancer regions), thereby turning genes on or off. Their DNA-binding specificity can be influenced by DNA sequence motifs and by epigenetic modifications. The core purpose of transcription factors is to ensure that genes are expressed in the right cells, at the right time, and at the right level throughout an organism’s life. TFs can work alone or as coordinated groups, recruiting or blocking RNA polymerase and other transcription machinery to regulate transcription positively (activation) or negatively (repression). A defining structural feature of TFs is that they contain at least one DNA-binding domain, which enables them to attach to regulatory DNA; proteins that help regulate transcription but lack DNA-binding domains (such as coactivators and chromatin remodelers) are not classified as transcription factors.
